A Hatters reserve side including 12 members of the club's youth teams came from a goal down to beat their Southend United counterparts at Roots Hall.
The 2-1 victory came thanks to two goals from Jack Wood and extends Alan Neilson and Kevin Watson's unbeaten record in the totesport.com Combination to nine games.
With three games left in the competition, it puts the Town's second string in line to lift the East Division title.
Southend fielded a strong line-up with Leicester City's Lee Cook on trial and striker James Walker making his comeback from injury in attack.
Cook created the opening goal on 25 minutes when his right-wing cross was headed home by Jamie Forshaw to give Southend the lead.
Back came the Town, however, and they were back on level terms when Wood scrambled home the loose ball from close range.
Wood then doubled his tally for the match seven minutes from time to give the young Hatters all three points when he was on hand to tap in again from close range after home goalkeeper Clark Masters had parried Taylor Nathaniel's shot.
The Town's reserves go in search of a 10th game unbeaten when they travel to Leyton Orient this Thursday, the 28th, at Brisbane Road for a 2pm kick-off.
