Calvin Andrew grabbed his first goal of the season to force an FA Cup replay with Brentford next Tuesday night at Griffin Park.
The Hatters youngster, who replaced Paul Furlong after just four minutes, latched onto a Darren Currie low cross late in the second half.
Brentford had been the more superior side in the first half and took a deserved half-time lead through Charlie Ide's hooked shot from the edge of the area.
But despite Furlong's withdrawal and losing Jaroslaw Fojut through injury at the start of the second half, Luton managed to keep their FA Cup dreams alive.
Furlong was back in the starting line-up after recovering from a groin injury, but the gamble to start with the striker didn't pay off as he hobbled off early.
Richard Jackson kept his place in the side with Alan Goodall returning to the bench after completing his one match suspension.
Brentford, who had made three changes to the side which were beaten by Macclesfield in midweek, opened proceedings brightly and with just two minutes gone, Ide's floating free-kick was met by Adrian Pettigrew's towering header from ten yards that flew inches over the crossbar.
In front of a spacious Kenilworth Road crowd, Luton began nervously. But Andrew's arrival almost had an immediate impact when his flicked header on seven minutes released Edwards, who cracked a shot over the crossbar from 12-yards.
But it was the visitors who continued to enjoy the majority of possession with Lee Thorpe and winger Ricky Shakes both having efforts blocked.
Luton were then unlucky not to break the deadlock on 16 minutes when Steve Robinson's flicked header released Andrew, whose hopeful cross found the feet of David Bell. The Luton midfielder then cut across two defenders and drilled a low shot that was palmed away by Brentford goalkeeper Simon Brown's low dive to his left.
As Luton struggled to find any rhythm to their play, Brentford continued to look dangerous on the attack and it was little surprise when they opened the scoring on 20 minutes after Thorpe's flicked header saw Ide hook a shot over Brill from the edge of the area.
The Hatters should have equalised six minutes later when Spring's long range shot ricocheted into the path of Edwards, but the midfielder wasted a golden opportunity, squandering his chance from eight yards by missing the target.
Brentford, whose manager Terry Butcher ripped into his side after Tuesday's loss at Macclesfield, were certainly responding in a positive manner and Thorpe was denied getting amongst the scorers after being put through in the clear, by a timely tackle from Chris Coyne just after the half-hour.
Referee Andy D'Urso once again invoked the wrath of Luton supporters when, after Spring was forced to go off for stitches, the Essex official refused to the let midfielder back on the pitch from the tunnel, forcing him to run half way around the other side of the pitch.
But Luton were simple struggling to make a breakthrough as the long ball tactics continue to have little effect.
AT HALF TIME: LUTON 0 BRENTFORD 1
Hatters boss Kevin Blackwell had to make changes, and he did immediately at the start of the half as Drew Talbot replaced the ineffective Edwards.
But just two minutes into the second half, Luton were forced into using their third substitute.
After Glenn Poole tried his luck from distance, hitting a 30-yard shot wide of Brill's left post, Jaroslaw Fojut was down in a heap and quickly left on a stretcher, leaving Goodall to come on at left-back, Jackson moving to right-back and Chris Perry slotting in at centre-half.
Despite the enforced change, Luton looked more sharper at the start of the second half and Currie was unlucky on 58 minutes when his free-kick took a faint deflection, veering his effort wide of the goal.
Currie then went even closer seven minutes later after Robinson bravely won a 50/50 challenge and when the ball broke to the former Ipswich man, his swerving effort from 30-yards missed the target by inches.
Brentford had been relatively quiet in the second half, but came close to adding a second when Sammy Moore was able to turn Goodall with ease, but his left-footed shot fizzed wide of Brill's right post.
Currie then had an acute shot blocked by Brown at his near post as Luton slowly began to throw men forward.
But it was lack of ideas that seemed to be Luton's downfall as hopeful balls up field failed to reach there intended target.
With time fast running out, Luton began to take a few chances and when it looked like an equaliser would never arrive, Andrew popped up with the goal to break Brentford hearts.
On 79 minutes, Currie drilled across a low cross from the right, which was turned home by the young Hatters striker from six-yards, under the body of Brown.
Luton had chances to snatch the win in the final moments of the game with Bell testing Brown, but Talbot went even closer after he failed to connect with an overhead kick after Coyne's rising header at the back post.
